Structure and Function of Membrane Proteins and Halophilic Proteins

The structure of several proteins has been determined by X-ray crystallography as well as by NMR techniques. One major focus is on the structure of membrane proteins like bacteriorhodopsin. Another aspect is the structure of halophilic proteins.

Bacteriorhodopsin

We have determined the structure of bacteriorhodopsin, which has become a paradigm for membrane proteins in general and transporters in particular.

Halorhodopsin

We have determined the structure of the light-driven chloride pump halorhodopsin.

Dodecin

Crystals of dodecin were obtained by shotgun crystallization directly out of the cytosol. Upon structure determination, an interesting riboflavin-binding protein was found.

Ferredoxin from Halobacterium

The structure of a 2Fe-2S ferredoxin with high similarity of the cyanobacterial protein was determined by NMR techniques. It revealed an acidic cluster which may represent an example of rapid adaptation after horizontal gene transfer.

Ferritin from Halobacterium

The structure of the Halobacterium ferritin was determined by X-ray crystallography.
