Using a recently in our research group established actin marker Lifeact-GFP my project aims to reinvestigate the organization and dynamics of actin in several established mammalian cell culture models. The goal of the project is to answer the questions how mechanical forces are transmitted throughout mammalian cells and how the cortical actomyosin network and its associated proteins react to external mechanical stimuli. For tackling these questions, I combine cell biological and biophysical methods to study the molecular composition, biophysical properties and biological functions of the cortical actomyosin network. In the final, I want to establish a biophysical model system of the overall force distributions at the cortical side of epithelial cells.
